KASP genotyping technology is based on competitive allele-specific PCR and enables bi-allelic scoring of single nucleotide polymorphisms (SNPs) and insertions and deletions (Indels) at specific loci.
KASP is easy to use – simply add your SNP-specific KASP Assay Mix and the universal KASP-TF Master Mix to your DNA samples, run a thermal cycling program, and measure the end-point fluorescence.
The allele-specific primers each harbour a unique tail sequence that corresponds with a universal FRET (fluorescence resonant energy transfer) cassette; one labelled with FAM™ dye and the other with HEX™ dye. The KASP-TF Master Mix contains the universal FRET cassettes, ROX™ passive reference dye, taq polymerase, free nucleotides and MgCl2 in an optimised buffer solution.
A key feature of our PCR-based KASP technology is the use of a universal FRET cassette reporter system that eliminates the need for costly dual-labelled probes. Our allele-specific forward primers each have a proprietary tail sequence that corresponds with one of two FRET cassettes: one label with FAM dye and the other with HEX dye. This technology gives KASP a clear cost advantage, and when combined with miniaturised reaction volumes (10 µL in 96-well PCR plates, 5 µL in 384-well PCR plates; 1 µL in 1536-well PCR plates), delivers an exceptionally accurate, flexible, and cost-effective genotyping solution. Bi-allelic discrimination is achieved through the competitive binding of the two allele-specific forward primers.
